This is possible, and a hacky method exists, but we don't recommend using it for a number of reasons. (Just search the forum for %updatepath%, for example).
That being said, we're working on adding standalone patches now. That is building a tiny standalone exe that requires no internet connection (or additional requirements) and can update your app.

We've been shifting around features. We're delaying standalone patches to (at least) wyBuild 2.8. The reason we shifted features like this is that a full installer is a "good enough" alternative to standalone patch. And we plan to add full installer ability to wyBuild 2.7.
